我们的实验室研究皮肤适应性进化的机理,尤其重点研究表皮屏障的形成方式。皮肤可分为表皮、真皮和皮下组织三层。表皮是由多层组成的复层鳞状上皮,其最外层是由死细胞组成的角质层(角质化层)。这层角质层是在古代两栖动物从水中向陆地过渡阶段进化而来的,它是陆生脊椎动物在陆地上生存的关键屏障。角质层是由一种特殊类型的细胞死亡形成的,被称为角质细胞凋亡,发生于颗粒层最顶部的SG1细胞中。我们的实验室已经开发了一种活细胞成像系统,可利用光学显微镜观察和了解角质细胞凋亡过程。

表皮活细胞成像面临哪些挑战?
鉴于我们研究的表皮具有多层结构,我们需要以三维方式对其结构进行成像。常规荧光显微镜和相机无法清晰地捕捉到这种3D结构,我们必须使用共聚焦显微镜。但共聚焦显微镜价格昂贵,因此我们的实验室难以购置一台。以前,我们不得不依赖于研究所和大学的共享设备。
我们的实验需要利用延时成像来观察颗粒层细胞的角质细胞凋亡过程,有时需要连续几天观察才能成像。但共享设备采用的是预约制,且使用时间有限。此外,每次成像都需要花费很长时间进行设置,所以我们需要一个这样孵化器,不仅可以培养细胞,还必须配置多点延时成像的设置。
鉴于以上情况,我们希望自己的实验室能够安装共聚焦显微镜,以便我们做实验时不受时间限制,也无须重复设置程序。
是什么让您决定引进MAICO的?
我们一直在使用滨松光子学株式会社的sCMOS相机进行荧光成像。在我们寻找共聚焦显微镜的过程中,一位销售代表向我们介绍了新发布的MAICO MEMS共聚焦装置,据说只需添加该装置即可将现有显微镜转变为共聚焦显微镜。我们首先要求当场演示。
虽然该装置价格很合理,但最初我很担心它的成像质量和灵敏度,我记得在演示过程中我非常惊讶,因为它竟然可以捕捉到不逊于其他公司共聚焦显微镜的高质量图像。基于以下几个因素,我们最终决定引进该装置:价格合理、成像质量与其他共聚焦显微镜相当、可以从一个波长开始,然后通过其模块化亚基结构添加更多波长,以及小型化的设计。
另一个决定性因素则是MAICO的独特功能可同时进行多波长观察,且无串扰。我们的实验室要进行活细胞成像,所以我们希望同时捕获多个波长。这往往导致波长泄漏问题,但MAICO解决了这个问题,对我们的工作大有裨益。
最近,除MAICO MEMS共聚焦装置外,我们还完成了多点延时成像用XY工作台和保持焦点位置用Z漂移补偿器的安装。现在我们终于有了共聚焦延时成像的完整设置,我们即将开始实际成像操作。

您如何看待MAICO的可用性?
我发现它非常人性化,操作也很简单。例如,针孔设置虽然无法进行精细调整,但可以分为S、M、L三个级别进行设置,简单易懂。如此一来,就能够很好地向每年新加入实验室的新生解释其操作过程了。此外,为了使每个镜头均达到上佳成像条件,滨松光子学株式会社提供了分辨率模拟器,我发现这个模拟器非常有用,让我们可以根据样品结构和厚度调整Z切片的厚度,同时参考模拟结果。
能否谈谈未来的研究展望?
目前,我们主要利用人类和两栖动物的表皮样品进行实验。未来,我们希望观察和研究两栖动物和爬行动物的表皮,以研究物种特异性差异与进化机理。我们计划将共聚焦显微镜图像与电子显微镜分析和遗传信息相结合,以开展更加详细的调查研究。
鉴于我们的实验室属于皮肤病学研究领域,有时我们会与皮肤科医生和化妆品制造商合作开展各种研究项目,包括特应性皮炎(皮肤屏障受损)研究与化妆品成分评估。我目前正在开发阐明表皮屏障形成机理用活细胞成像系统,该系统也有望用于这些研究。因此,我们同时致力于开发新的评估系统/仪器。
